Efficiency here is distance per unit of fuel — km per litre or miles per gallon — not litres per 100 km, which is its reciprocal. Mixing the two is the commonest error in fuel calculations and produces an answer wrong by a large factor rather than a small one. Fuel is also only part of the cost of driving: depreciation, tyres and servicing typically add more again per mile.

How do I calculate fuel cost for a trip?
Divide the distance by the fuel efficiency to get the fuel needed, then multiply by the price. 300 km at 15 km/L needs 20 litres.
Should I use km/L or L/100km?
Either, but not interchangeably — they are reciprocals. This page expects distance per unit of fuel; if your figure is litres per 100 km, convert it first.
Is fuel the real cost of driving?
Only part of it. Depreciation, tyres, servicing and insurance typically add more again per mile, which is why reimbursement rates are well above fuel alone.
